5.1 This plate format is useful for the routine monitoring of culturable, waterborne bacteria in potable and non-potable waters. The significance of finding these bacteria can help with identifying water quality or water system problems or evaluate compliance with maintenance protocols. This test method uses small volumes of water, or dilutions thereof, and provides an easy and reliable method that eliminates media preparation and reduces laboratory waste.1.1 This test method describes a simple procedure for the quantification of culturable, waterborne bacteria in potable water (drinking water, bottled water, and dental water, for example) and non-potable waters (cooling towers, for example).1.1.1 The EasyDisc2, 3 plate format is designed to test 1 mL of a water sample on a 47 mm gridded plate containing a growth reagent embedded to the plate’s inner surface.1.1.2 Detection is based on colorimetric technology in which viable, aerobic, heterotrophic, waterborne bacteria grow when present in the water sample, displaying a color reaction which allows for a simplified visualization of colony growth.1.2 Each plate can accurately detect up to 300 colony forming units per 1 mL (CFU/1 mL) of sample. To increase the quantification range, a sample dilution can be used. Adjust the CFU/mL result to reflect dilutions.1.3 This test method can be used for potable (for example, drinking, bottled, and dental) waters and non-potable waters such as cooling tower waters. It is the user’s responsibility to adhere to all requirements by local regulations and ensure the validity of this test method for waters other than those tested as part of the Interlaboratory Study (ILS).1.4 The values stated in SI units are to be regarded as the standard. 5.1 Sounding tubes may be fabricated from 11/2 NPS or larger. Only when otherwise specified, Schedule 40 components, manufactured from the list of material indicated in Practice F1155 and Specification A53/A53M, Grade S or Grade ERW. In addition, sounding tubes may be fabricated in stainless steel for stainless steel tanks.5.1.1 Sounding tubes passing through or terminating in fuel tanks, potable water tanks, or clean salt water ballast tanks should be constructed of 70-30 copper nickel, but other suitable material is acceptable.5.2 Striker plates shall be fabricated in accordance with Specification A36/A36M.5.3 The fittings shall be designed in accordance with ASME B16.5, ASME B16.9, ASME B16.28, or ASME B16.11 as applicable (see Table 21 in Practice F1155), and the installation shall be in accordance with ASME B31.1 as modified by Specification F722. These standards cover the fitting tolerances.5.4 Some cargo may preclude the use of materials specified in this guide. However, configuration examples are applicable for all applications.5.5 When a sounding tube is combined with the air escape, either three 11/4-in. (approximately 30-mm) diameter holes approximately 12 in. (305 mm) from the tank top equally spaced or six 1/2-in. (approximately 15-mm) diameter holes approximately 6 in. (150 mm) from the tank top equally spaced can be used for perforations. This guide covers standard specification for precipitation-hardening stainless steel plate, sheet, and strip. The mechanical properties of these steels shall be developed by suitable low-temperature heat treatments or precipitation hardening. The steel shall conform to the required chemical composition for carbon, manganese, phosphorus, sulfur, silicon, chromium, nickel, aluminum, molybdenum, titanium, copper, and other elements. Material of types other than Type UNS S46910 shall be furnished in solution-annealed condition while Type UNS S46910 shall be furnished in cold-worked condition. The steel shall conform to the required mechanical properties such as tensile strength, yield strength, elongation, and hardness. Samples cut from the solution-annealed plate, sheet, or strip shall withstand cold bending without cracking on the outside of the bent portion. Samples cut from the solution-annealed plate, sheet, or strip shall conform to the specified mechanical properties when precipitation hardened.1.1 This specification2 covers precipitation-hardening stainless steel plate, sheet, and strip. The mechanical properties of these steels are developed by suitable low-temperature heat treatments generally referred to as precipitation hardening.1.2 These steels are used for parts requiring corrosion resistance and high strength at room temperature or at temperatures up to 600 °F (315 °C). Some of these steels are particularly suitable for moderate to severe drawing and forming in the solution-treated condition. Others are capable of mild forming only. This specification covers plate consisting of a base metal to which is bonded, integrally and continuously, on one or both sides a layer of one of the following: titanium, zirconium, tantalum, niobium, and their alloys. The material generally is intended for pressure vessel use. The cladding metal shall be bonded to the base metal by any cladding operation that will produce a clad product which will conform to the requirements of this specification. When the cladding operation does not affect the chemical composition of the cladding or base metal, or both, the chemical analysis, in accordance with cladding, or base metal specifications or both, may be performed prior to the cladding operation. All heat treatments shall be performed as needed in the cladding operation unless otherwise specified or agreed between the purchaser and the manufacturer. Mechanical testing of the base metal, in accordance with the base metal specification, may be performed prior to the cladding operation. The tensile properties shall be determined by a tension test on the base metal only in accordance with the testing requirements of the base metal specification. The specimen orientation, location with respect to thickness, and number of tests and retests shall be in accordance with the requirements of the base metal specification when tension, bend, and impact tests shall be performed. 5.1 The glass petri plate provides a closed system for enumeration and easy application of a pre-saturated or impregnated antimicrobial towelette by an analyst.5.2 Inoculation of carriers (five 10 µL spots of microbial suspension) is conducted using a template and a positive displacement pipette, thereby ensuring a precise inoculum level and uniform distribution of inoculum.5.3 A single towelette is tested per carrier, thereby ensuring comparable treatment among carriers and eliminating the likelihood of cross-contamination between carriers.5.4 The circular motion of the product application (wipe outside to inside, lift towelette to invert and wipe inside to outside) is a relevant motion that ensures uniform coverage and contact of disinfectant with the inoculated surface.5.5 The addition of neutralizer to the treated plates ensures thorough neutralization at the end of the product’s contact time. This test method provides a procedure for performing neutralization verification to confirm that the microbicidal and/or microbistatic activity of a test substance has been brought to an undetectable level at the end of the contact time.5.6 The design of the test method minimizes any loss of viable organisms through carrier wash-off.5.7 This test method provides for optional use of an organic soil load as dictated by a product’s label claim.5.8 It is optional to adjust (either dilute or concentrate) the inoculum level to achieve desired control carrier counts and to accommodate different product performance standards.1.1 This test method provides detailed instructions for performing a quantitative evaluation of antimicrobial efficacy of a towelette when challenged against Staphylococcus aureus, Pseudomonas aeruginosa and Salmonella enterica. The method may be used with other microbial strains, though modification may be necessary to accommodate recovery.1.1.1 Antimicrobial towelettes, designed to decontaminate hard, non-porous surfaces, are diverse in size, matrix composition, and packaging.1.1.2 Antimicrobial towelettes also vary in label claims and use directions.1.2 This quantitative method does not differentiate between mechanical removal of inoculum from a surface and chemical inactivation of the test microbe; rather, product efficacy is considered a combination of both attributes of a towelette-based formulation.1.3 It is the responsibility of the investigator to determine whether Good Laboratory Practices (GLP Standards—40 CFR, Part 160 of FIFRA) are required and to follow them when appropriate.1.4 This standard may involve the use of hazardous materials, chemicals and infectious microorganisms and should be performed only by persons with formal training in microbiology.1.5 Strict adherence to the protocol is necessary for the validity of the test results.1.6 The values stated in SI units are to be regarded as standard.
