This guide is intended for use in any laboratory utilizing PCR or RT-PCR to amplify and detect nucleic acid sequences of mycobacteria from a biological preparation and to identify the species of origin.The criteria used for the identification and evaluation of the amplification reactions should be administered by an individual trained in the use of molecular biological and microbiological techniques associated with PCR and MTB.1.1 This guide covers basic considerations, criteria, principles and recommendations that should be helpful when developing, utilizing, or assessing PCR-specific protocols for the amplification and detection or identification of mycobacterial nucleic acids. This guide is not a specific protocol for the detection of specific mycobacteria. It is intended to provide information that will assist the user in obtaining high quality and reliable data. The guide is closely related to and should be used concurrently with the general PCR Guide E 1873.1.2 This guide has been developed for use in any molecular biology or biotechnology laboratory. It may be useful for the detection of mycobacteria in clinical, diagnostic laboratories.1.3 This guide does not cover details of the various methods such as gel electrophoresis that can be utilized to help identify PCR-amplified mycobacterial nucleic acid sequences, and it does not cover details of instrument calibration.1.4 This guide does not cover specific variations of the basic PCR or RT-PCR technology (for example, quantitative PCR, multiplex PCR and in situ PCR), and it does not cover details of instrument calibration.1.5 Warning-Laboratory work involving certain clinical specimens and microorganisms can be hazardous to personnel. Precaution: Biosafety Level 2 facilities are recommended for potentially hazardous work, and Biosafety Level 3 facilities are required for propagating and manipulating Mycobacteria tuberculosis cultures (). This specification covers polyvinyl chloride and other conforming organic polymer-coated steel chain-link fabric, coated before weaving. The intended use of this practice is for chain link fencing of varying heights and designs to be used to enclose a sports field, sport court or recreation facility including the internal fencing required for safety, separation of activities, security, crowd control, access and egress or other requirements.Consideration should be given to fence offset distances from the activity field to provide a safety area for the participants and viewers.This practice is not intended for applications where fencing higher than 12 ft (3660 mm) is desired by the owner.Follow Guide F 1553 format to specify the chain link fence material and installation.Warning Regarding Windscreens and Added Fence Padding—If windscreens or padding are to be installed at the time of fence erection or at a later time, it is advisable to use stronger framework, closer post spacing or back bracing of posts depending on the type of screening material to be used, area of the fence covered and the local wind and weather conditions. This specification deals with Grade 80 heat-treated alloy steel chain for applications such as slings, lifting assemblies, and load binding. The alloy steel chain shall be made to a fully-killed fine austenitic grain process. Alloy steel chain may be made by the electric welding or gas welding process. The material shall be heat treated which includes quenching and tempering. Material's composition shall conform to the chemical requirements for carbon, phosphorous, sulfur, nickel, chromium, and molybdenum. The material shall conform to the mechanical property requirements for breaking force and elongation and to the dimensional requirements for the appropriate size chain. All chain shall be tested to at least the proof load.1.1 This specification covers Grade 80 heat-treated alloy steel chain for such applications as slings, lifting assemblies, and load binding. For overhead lifting applications, only alloy chain should be used.1.2 The chain grade is based on the nominal stress in the link at the design breaking force strength. It is calculated by taking the minimum breaking force load and dividing by two times the nominal cross-sectional area of the link.1.3 The values stated in either SI units or in other units are to be regarded separately as standard. 5.1 The Intended Use of This Guide is to identify the specific functions and qualities desired of a pickleball fence and offer recommendations on how to achieve them with a chain link fence for different types of pickleball courts.5.2 The Primary Function of a Pickleball Court Fence is to keep pickleball balls in and uninvited traffic out. It also serves as a barrier to prevent players from colliding with fixed objects and spectators. It also prevents players from running onto irregular or slippery surfaces or into dangerous adjacent areas. Fences need to respond to secondary functional requirements, including: spectator viewing, screening the court from wind and visual distractions outside the court, and being as open and welcoming as security conditions permit. This specification covers zinc-coated (galvanized) steel chain-link fence fabric, zinc coated either before or after weaving. The wire shall be woven throughout in the form of approximately uniform square mesh, having parallel sides and horizontal and vertical diagonals of approximately uniform dimensions. This specification covers carbon steel chain for such applications as railroad cars, construction, industrial uses, load binding, and general purposes other than overhead lifting. The three classes of carbon steel chains covered here include Grade 30 (proof coil chain), Grade 43 (high test chain), and Grade 70 (transport chain). The dimensional requirements of the chain with respect to the appropriate grade and size are presented. The mechanical testing requirements for the steel includes, proof testing, breaking force testing, and elongation testing. One test for breaking strength and elongation shall be made from each lot. This specification prescribes the minimum requirements for the design, performance, testing, and certification of protective garments and protective devices that, when worn by operators of power chain saws, shall provide cut resistance protection to their legs, and thereby reducing leg injuries caused by close contact with the running chain saw. The protective garments and devices shall be constructed with suitable fit and flexibility so as to provide sufficient leg comfort and normal maneuverability to perform intended tasks, while maintaining satisfactory functionality and efficacy for the duration of its regular use throughout seasonal climatic variations.
